Search for fertile individuals of the river pearl mussel in Upper Malša in the project ATCZ37 Malsemuschel financed by the program Interreg - V-A Austria - Czech Republic

In order to strengthen the population of the river pearl (Margaritifera margaritifera (Linnaeus, 1758)) breeding is carried out as part of the rescue program for this species. Larvae, so-called glochidia, taken from wild mothers enter the rearing. It is the phase of glochidia collection that is critical for maintaining the genetic variability of the population. By sampling from a few individuals, sometimes even repeatedly from the same ones, only a small portion of the original genetic variability is transferred to the new generation. Loss of variability significantly increases the probability of population extinction. The subject of the order is the search for glochidia-producing individuals of the river pearl mussel in the Malše stream (in the vicinity of Dolní Dvořiště) during the breeding season of pearl mussel (i.e. in August and September) in the seasons 2019 and 2020. The work is a continuation of other activities in the "Malsemuschel" project (https://www.at-cz.eu/cz/ibox/po-2/atcz37_malsemuschel), in particular the mapping of occurrence pearls and water temperature monitoring. The work will be implemented in 2 parts. The first part will be handed over to the customer by September 30, 2019, and the second by September 30, 2020. The order is divided into parts according to the year of implementation, both parts have the same scope of work. Based on these, he searches for at least 20 individuals identifiable as females before the start of reproduction and marks them. These individuals must cover the detected occurrence of mother-of-pearl in Malša as representatively as possible. In these individuals, glochidia production will be monitored until their release or the end of the breeding season. The exact date varies from year to year and depends on the weather. Monitoring the development of glochidia will be carried out by taking water from the ejection hole every other day for the entire period of their maturation until they are ejected. The collected sample will be checked on the spot under a microscope for the presence of glochidia and their stage of development. The development of glochidia is controlled by temperature, so data from continuous temperature measurement in the flow will be available to the contractor.
